RESUMO

The foregut, located at the front of the digestive tract, serves a vital role in insects by storing and grinding food into small particles. The innermost layer of the foregut known as the chitinous intima, comes into direct contact with the food and acts as a protective barrier against abrasive particles. Knickkopf (Knk) is required for chitin organization in the chitinous exoskeleton, tracheae and wings. Despite its significance, little is known about the biological function of Knk in the foregut. In this study, we found that LmKnk was stably expressed in the foregut, and highly expressed before molting in Locusta migratoria. To ascertain the biological function of LmKnk in the foregut, we synthesized specific double-stranded LmKnk (dsLmKnk) and injected it into locusts. Our findings showed a significant decrease in the foregut size, along with reduced food intake and accumulation of residues in the foregut after dsLmKnk injection. Morphological observations revealed that newly formed intima became thinner and lacked chitin lamella. Furthermore, fluorescence immunohistochemistry revealed that LmKnk was located in the apical region of new intima and epithelial cells. Taken together, this study provides insights into the biological function of LmKnk in the foregut, and identifies the potential target gene for exploring biological pest management strategies.

We report compound heterozygous variants in TOE1 in siblings of Chinese origin who presented with dyskinesia and intellectual disabilities. Our report provides further information regarding the etiology and pathogenesis of pontocerebellar hypoplasia type 7 syndrome (PCH7). Clinical manifestations were obtained, and genomic DNA was collected from family members. Whole-exome and Sanger sequencing were performed to identify associated genetic variants. Bioinformatics analysis was conducted to identify and characterize the pathogenicity of the heterozygous variants. Following long-term rehabilitation, both siblings showed minimal improvement, and their condition tended to progress. Whole-exome sequencing revealed two unreported heterozygous variants, NM_025077: c.C553T (p.R185W) and NM_025077: c.G562T (p.V188L), in the TOE1 gene mapped to 1p34.1. Sanger sequencing confirmed that the two variants in the proband and her brother were inherited from their parents. The NM_025077: c.C553T (p.R185W) variant was inherited from the father, and the NM_025077: c.G562T (p.V188L) variant was inherited from the mother. Although the two variants in the TOE1 gene have not been reported previously, they were associated with PCH7 based on integrated analysis. Thus, our report contributes to our knowledge regarding the etiology and phenotype of PCH 7.

Although membrane separation technology has been widely used in the treatment of oily wastewater, the complexity and high cost of the membrane preparation, as well as its poor stability, limit its further development. In this study, via the vacuum-assisted suction filtration method, polydopamine (PDA)-coated TiO2 nanoparticles were tightly attached and embedded on both sides of laboratory filter paper (FP). The resultant FP possessed the typical wettability of high hydrophilicity in the air with the water contact angle (WCA) of 28°, superoleophilicity with the oil contact angle (OCA) close to 0°, underwater superoleophobicity with the underwater OCA greater than 150°, and superhydrophobicity under the water with the underoil WCA over 150° for five kinds of organic solvents (carbon tetrachloride, toluene, n-hexane, n-octane, and iso-octane). The separation efficiency of immiscible oil/water, oil-in-water, and water-in-oil emulsions using the modified FP is higher than 99%. After 17 cycles of emulsion separation, a high separation efficiency of 99% was still maintained for the FP, along with good chemical and mechanical stability. In addition, successful separation and purification were also realized for the oil-in-water emulsion that contained the methylene blue (MB) dye, along with the complete degradation of MB in an aqueous solution under UV irradiation.

The rice cytochrome P450 gene has been comprehensively studied in the present study. This gene encodes CYP90D5 in promoting the degradation of isoproturon (IPU) and acetochlor (ACT) in rice tissues and grains. It has here been found that CYP90D5 improved the resistance of the plant to IPU and ACT, which was reflected in the improvement of the growth of the overexpression (OE) lines. CYP90D5 also reduced the levels of IPU and ACT accumulation in rice, and the CRISPR-Cas9 (Cas9) lines displayed the opposite effects. This function of CYP90D5 for pesticide degradation was also confirmed by the transformation of CYP90D5 in Pichia pastoris. Compared with the control yeast, it grew better and could degrade more pesticides. In addition, the relative contents of the IPU and ACT derivatives increased in the OE rice, while they decreased in the Cas9 rice. This suggested that CYP90D5 plays a pivotal role in the pesticide detoxification and degradation.

The design and fabrication of advanced membrane materials for versatile oil/water separation are major challenges. In this work, a superwetting stainless steel mesh (SSM) modified with in situ-grown TiO2 was successfully prepared via one-pot hydrothermal synthesis at 180 °C for 24 h. The modified SSM was characterized by means of scanning electron microscopy, energy spectroscopy, and X-ray photoelectron spectroscopy analysis. The resultant SSM membrane was superhydrophilic/superoleophilic in air, superoleophobic underwater, with an oil contact angle (OCA) underwater of over 150°, and superhydrophobic under oil, with a water contact angle (WCA) as high as 158°. Facile separation of immiscible light oil/water and heavy oil/water was carried out using the prewetting method with water and oil, respectively. For both "oil-blocking" and "water-blocking" membranes, the separation efficiency was greater than 98%. Also, these SSMs wrapped in TiO2 nanoparticles broke emulsions well, separating oil-in-water and oil-in-water emulsions with an efficiency greater than 99.0%. The as-prepared superwetting materials provided a satisfactory solution for the complicated or versatile oil/water separation.

Robust membrane materials with high efficiency have attracted extensive attention in oil/water separation. In this work, carbon particles via candle combustion were firstly adsorbed on the surface of stainless steel meshes (SSMs), which formed a thin hydrophobic coating, and a rough structure was then constructed through chemical vapor deposition and high temperature calcination, with the resultant SSM surface wrapped with uniform silica coating possessing the characteristic of superoleophobicity underwater. Scanning electron microscope (SEM), energy dispersive spectroscopy (EDS), and X-ray powder diffraction (XRD) were used to characterize the modified SSMs. The prepared SSMs were superhydrophilic in air, and they had superoleophobicity underwater (157.4°). The separation efficiency of five oil/water mixtures was above 98.8%, and the separation flux was 46,300 L·m-2·h-1. After it was immersed in 1 mol/L NaOH, 1 mol/L HCl and 3.5 wt% NaCl for 24 h, respectively, the efficiency was still above 97.3%. Further immersion in the solution of dopamine and octadecylamine resulted in the transformation of superhydrophililc/superoleophobicity-underwater SSMs to superhydrophobic SSMs, and the resultant SSMs with reverse surface wettability was also used for the oil/water separation with good separation efficiency and separation flux.

PURPOSE: To observe the efficacy and safety of intranasal dexmedetomidine combined with midazolam in cranial magnetic resonance imaging of children. DESIGN: A prospective, observational, single-arm, one-center study. METHODS: A total of 474 children were scheduled for cranial 3.0 T MRI at the first time. All patients were initially given 3 mcg/kg dexmedetomidine combined with 0.15 mg/kg midazolam. The one-time success rate, vital signs before and after treatment, onset time, recovery time, and incidence of adverse reactions were recorded. FINDINGS: The one-time success rate was 78.1%. There were significant differences in respiration, heart rate, and blood oxygen saturation before and after treatment (P < .001). The onset time was 10 (8-15) minutes. The average recovery time was 2.58 ± 1.10 hours. Only 1.27% (6 cases) of adverse reactions were observed, including bradycardia (3 cases, 0.6%), tachycardia (1 case, 0.2%), and startle (2 cases, 0.4%). No special treatment was needed. The success of the examination was significantly correlated with age (OR 1.320, 95% CI 1.019-1.710, P = .035) and onset time (OR 0.959, 95% CI 0.921-0.998, P = .038). CONCLUSION: Dexmedetomidine 3 mcg/kg combined with midazolam 0.15 mg/kg intranasally has a good sedative effect in pediatric cranial magnetic resonance examinations, little impact on breathing and circulation, and few adverse reactions. Age and onset time are related factors affecting the one-time success rate.

Fall armyworm, Spodoptera frugiperda (J. E. Smith), has become an important agricultural pest worldwide. S. frugiperda is mainly controlled by the chemical insecticides, whereas the frequent application of insecticides would result in the resistance development. Insect uridine diphosphate-glucuronosyltransferases (UGTs), as phase II metabolism enzymes, play vital roles in the breakdown of endobiotic and xenobiotics. In this study, 42 UGT genes were identified by RNA-seq, including 29 UGT genes were elevated compared to the susceptible population, and the transcript levels of 3 UGTs (UGT40F20, UGT40R18, and UGT40D17) were increased by more than 2.0-fold in the field populations. Expression pattern analysis revealed that S. frugiperda UGT40F20, UGT40R18, and UGT40D17 were increased by 6.34-, 4.26-, and 8.28-fold, compared the susceptible populations, respectively. The expression of UGT40D17, UGT40F20, and UGT40R18 was affected after exposure to phenobarbital, chlorpyrifos, chlorfenapyr, sulfinpyrazone, and 5-nitrouracil. The induced expression of UGT genes may have improved UGT enzymatic activity, while the inhibition of UGTs genes expression may decreased UGT enzymatic activity. Sulfinpyrazone, and 5-nitrouracil, significantly increased the toxicity of chlorpyrifos and chlorfenapyr, as well as phenobarbital significantly reduced the toxicity of chlorpyrifos and chlorfenapyr against the susceptible populations and field populations of S. frugiperda. The suppression of UGTs (UGT40D17, UGT40F20, and UGT40R18) significantly increased the insensitivity of the field populations to chlorpyrifos and chlorfenapyr. These findings strongly supported our viewpoint that UGTs may play a critical role in insecticide detoxification. This study provides a scientific basis for the management of S. frugiperda.

Construction of different surface wettability is meaningful for the interaction between the sorbent surface and target components. In the current study, four kinds of stainless-steel wires (SSWs) with different hydrophobic/hydrophilic property were prepared and used as the absorbents to enrich the target compounds with different polarity. Comparative extraction of six non-polar polycyclic aromatic hydrocarbons (PAHs) and six polar estrogens was carried out by in-tube solid phase microextraction (IT-SPME). The results showed that two SSWs with the superhydrophobic surfaces exhibited high extraction capacity to the non-polar PAHs with the superior enrichment factor (EF) in the range of 29-672 and 57-744, respectively. In contrast, the superhydrophilic SSWs demonstrated higher enrichment efficiency for the polar estrogens than other hydrophobic SSWs. On the basis of optimized conditions, a validated analysis method was established using six PAHs as model analytes for IT-SPME-HPLC. Acceptable linear ranges (0.5-10 µg L-1) and low detection limits (0.0056-0.32 µg L-1) were achieved using the superhydrophobic wire modified by perfluorooctyl trichlorosilane (FOTS). The relative recoveries spiked at 2, 5 and 10 µg L-1 in the lake water samples were in the range of 81.5%-113.7%. The relative standard deviation (RSD) of intraday (≤0.8%, n = 3) and interday (≤5.3%, n = 3) tests demonstrated the good extraction repeatability for the same extraction tube. Satisfactory repeatability for the preparation of extraction tubes (n = 3) was also obtained with the RSD values in the range of 3.6%-8.0%.

The aim of this study was to prepare oridonin liposomes and evaluate the physicochemical characteristics and pharmacokinetics in rats. A three-level, three-factor Box-Behnken design was used to optimize the preparation of oridonin liposomes. A highly sensitive high-performance liquid chromatographic quantification method using ultraviolet detection was established and validated for the determination of oridonin in rat plasma. Twelve Sprague-Dawley rats were randomly assigned and injected with 15 mg/kg of oridonin or oridonin liposomes via the tail vein. Pharmacokinetic parameters were estimated using a compartmental modeling approach using PKsolver software. The optimum conditions were as follows: soybean phospholipids/cholesterol ratio, 3.9:1; soybean phospholipids/drug ratio, 8.5:1; and soybean phospholipid concentration, 1.1%. Under these conditions, the mean particle size, polydispersity index, zeta potential, and encapsulation efficiency of oridonin liposomes were 170.5 nm, 0.246, -30.3 mV, and 76.15%, respectively. The pharmacokinetic results showed that liposomes could significantly prolong the elimination half-life (from 2.88 ± 0.55 to 13.67 ± 3.52 h), increase the area under the concentration-time curve (from 1.65 ± 0.17 to 6.22 ± 0.83 µg h/ml), and decrease the clearance (from 6.62 ± 1.38 to 1.96 ± 0.24 L/kg h). The oridonin liposomes increased the elimination half-life and area under the concentration-time curve and provided a reference for the development of drugs with a short half-life.

BACKGROUND: Swallowing disorder is a common clinical symptom that can lead to a series of complications, including aspiration, aspiration pneumonia, and malnutrition. This study aimed to investigate risk factors of post-extubation dysphagia (PED) in intensive care unit (ICU) patients with endotracheal intubation, and to develop a risk-predictive model for PED, which could serve as an assessment tool for the prevention and control of PED. METHODS: Patients retrospectively selected from June to December 2021 in a tertiary hospital served as the derivation cohort. Patients recruited from the same hospital from March to June 2022 served as the external validation cohort for the predictive model. We used a combination of variable screening and least absolute shrinkage and selection operator (LASSO) regression to select the most useful candidate predictors and checked the multicollinearity of independent variables using the variance inflation factor method. Multivariate logistic regression analysis was performed to calculate the odds ratio (OR; 95% confidence interval [95% CI]) and P-value for each variable to predict diagnosis. The screened risk factors were introduced into R software to build a nomogram model. The performance of the model, including discrimination ability, calibration, and clinical benefit, was evaluated by plotting the receiver operating characteristic (ROC), calibration, and decision curves. RESULTS: A total of 305 patients were included in this study. Among them, 235 patients (53 PED vs. 182 non-PED) were enrolled in the derivation cohort, while 70 patients (17 PED vs. 53 non-PED) were enrolled in the validation cohort. The independent predictors included age, pause of sedatives, level of consciousness, activities of daily living (ADL) score, nasogastric tube, sore throat, and voice disorder. These predictors were used to establish the predictive nomogram model. The model demonstrated good discriminative ability, and the area under the ROC curve (AUC) was 0.945 (95% CI 0.904-0.970). Applying the predictive model to the validation cohort demonstrated good discrimination with an AUC of 0.907 (95% CI 0.831-0.983) and good calibration. The decision-curve analysis of this nomogram showed a net benefit of the model. CONCLUSION: A predictive model that incorporates age, pause of sedatives, level of consciousness, ADL score, nasogastric tube, sore throat, and voice disorder may have the potential to predict PED in ICU patients.

BACKGROUND: DNA replication alteration is a hallmark of patients with lung adenocarcinoma (LUAD) and is frequently observed in LUAD progression. Origin recognition complex (ORC) 1, ORC2, ORC3, ORC4, ORC5, and ORC6 form a replication-initiator complex to mediate DNA replication, which plays a key role in carcinogenesis, while their roles in LUAD remain poorly understood. METHODS: The mRNA and protein expression of ORCs was confirmed by the GEPIA, HPA, CPTAC, and TCGA databases. The protein-protein interaction network was analyzed by the GeneMANIA database. Functional enrichment was confirmed by the Metascape database. The effects of ORCs on immune infiltration were validated by the TIMER database. The prognostic significance of ORCs in LUAD was confirmed by the KM-plot and GENT2 databases. DNA alteration and protein structure were determined in the cBioProtal and PDB databases. Moreover, the protein expression and prognostic value of ORCs were confirmed in our LUAD data sets by immunohistochemistry (IHC) staining. RESULTS: ORC mRNA and protein were significantly increased in patients with LUAD compared with corresponding normal tissue samples. The results of IHC staining analysis were similar result to those of the above bioinformatics analysis. Furthermore, ORC1 and ORC6 had significant prognostic values for LUAD patients. Furthermore, the ORC cooperatively promoted LUAD development by driving DNA replication, cellular senescence, and metabolic processes. CONCLUSION: The ORC, especially ORC1/6, has important prognostic and expression significance for LUAD patients.

Due to the higher birth rate of preterm infants and improvements in their management, metabolic bone disease of prematurity (MBDP) has a high incidence and is attracting attention. However, clear indicators for the early diagnosis of MBDP are lacking. We aimed to explore simple and feasible early warning indicators for diagnosing MBDP. Our study collected case data of premature infants from two medical centers in Chongqing from January 2020 to February 2022. According to the inclusion and exclusion criteria, data from 136 cases were collected. The correlation between 14 variables in each case and the occurrence of MBDP was analyzed. According to area under the receiver operating characteristic curve (AUROC) analysis, the best cutoff value for each variable was determined. Potential predictors were selected, and Least Absolute Shrinkage and Selection Operator (LASSO) regression analysis was used to establish the association of two models with MBDP, whose results were used to develop a diagnostic nomogram. Furthermore, a model decision curve was analyzed. Four predictors were selected from 14 clinical variables by LASSO regression, and Model I was established, including the following characteristics: height (>36 cm), head circumference (≤29.49 cm), total serum calcium (Ca) (>2.13 mmol/L), and alkaline phosphatase (ALP) (>344 U/L) levels. A single predictor, the ALP level (>344 U/L), was used to establish Model II. The AUROC values of the two models were 0.959 for Model I and 0.929 for Model II. In conclusion, in this study, two diagnostic models of MBDP were developed using four combinations of predictors and ALP as a single predictor. Both models showed good sensitivity and specificity for the early diagnosis of metabolic bone disease (MBD), and an ALP level of 344 U/L was defined as a simple and effective diagnostic threshold. In future studies, using larger samples, diagnostic threshold values of ALP for premature infants of different ages should be established, and internal and external validations are needed to improve the adaptability of the current model.

Severe hemorrhage-induced acute lung injury (ALI) remains the major contributor to critical patient mortality and is associated with posthemorrhagic shock mesenteric lymph (PHSML) return. Omega-3 polyunsaturated fatty acids (ω-3 PUFAs) play overall protection on acute hemorrhage, but a reliable mechanism needs to be identified. The aims of this study were to investigate the role of ω-3 PUFAs in alleviating ALI and whether is related to the endotoxin contained in PHSML. Mesenteric lymph was harvested from rats subjected to hemorrhagic shock (hemorrhage-induced hypotension of 40 ± 2 mmHg for 90 min plus by resuscitation) or sham shock. The effect of ω-3 PUFAs on pulmonary function, water content, morphology, and LBP, CD14, TNF-α, and IL-6 levels were observed in rats subjected to hemorrhagic shock, while the effect of PHSML intravenous infusion on the beneficial effect of ω-3 PUFAs also was investigated. In addition, the effect of ω-3 PUFAs on the endotoxin contents in mesenteric lymph were detected. Hemorrhagic shock-induced ALI was characterized by increased functional residual capacity (FRC), lung resistance (RI), inspiratory capacity (IC), respiratory frequency, water contents and structural damage, along with increases in LBP, IL-6, and TNF-α. ω-3 PUFAs treatment reduced FRC, RI, IC, frequency, water contents, LBP, IL-6, TNF-α, and alleviated morphological damage. In contrast, PHSML infusion abolished the advantageous effects of ω-3 PUFAs on the above indices and CD14. Furthermore, the endotoxin level of PHSML was significantly enhanced, but declined following ω-3 PUFAs administration. These findings together suggested that treatment with ω-3 PUFAs ameliorates hemorrhagic shock-induced ALI, which is associated with reduced endotoxin contained in PHSML.

Water/oil separation from their mixture and emulsion has been a prominent topic in fundamental research and in practical applications. In this work, a smart superhydrophobic membrane (SHP) was obtained by dipping an off-the-shelf laboratory filter paper in an ethanol suspension of trichloro (1H,1H,2H,2H-tridecafluoro-n-octyl) silane, tetraethyl orthosilicate, and titanium dioxide nanoparticles with different dimensions of 20 and 100 nm. The selection of membrane substrates was optimized including different quantitative and quantitative filter papers with different filtration velocity (slow, intermediate, and fast). The as-prepared SHP was demonstrated to be superhydrophobic and photosensitive, which was used in the separation of carbon tetrachloride and water from their mixture and emulsion. Moreover, orderly aligned micropores were formed for the modified superhydrophobic filter papers by using nanosecond laser. Unidirectional penetration was obtained for the UV-irradiated paper with a bored pore in the range of 50-500 µm in the systems of air/water and water/oil. This study may promote the understanding of unidirectional transportation of liquid droplet and facilitate the design of interfacial materials with Janus-type wettability.

A robust superhydrophobic brass mesh was fabricated based on a low-energy surface and a roughness on the nano/micro-meter scale. It was carried out by the forming of hydroxyapatite (HP) coatings on its surface through a constant current electro-deposition process, followed by immersion in fluoroalkylsilane solution. Surface morphology, composition and wetting behavior were investigated by field-emission scanning electron microscopy (SEM), X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), high speed camera, and contact angle goniometer. Under optimal conditions, the resulting brass mesh exhibited superhydrophobicity, excellent anti-corrosion (η = 91.2%), and anti-scaling properties. While the surfactant liquid droplets of tetradecyl trimethyl ammonium bromide (TTAB) with different concentration were dropped on the superhydrophobic surface, maximum droplet rebounding heights and different contact angles (CAs) were observed and measured from side-view imaging. The plots of surfactant-concentration-maximum bounding height/CA were constructed to determine its critical-micelle-concentration (CMC) value. Close CMC results of 1.91 and 2.32 mM based on the determination of maximum rebounding height and CAs were obtained. Compared with its theoretical value of 2.1 mM, the relative errors are 9% and 10%, respectively. This indicated that the novel application based on the maximum rebounding height could be an alternative approach for the CMC determination of other surfactants.

The application of straw biochar in farmland in Dongting Lake area can realize the resource utilization of straw and reduce environmental risk. In 2020, a rice pot experiment was conducted to investigate the effects of different biochar application levels on ammonia volatilization rate, cumulative ammonia volatilization, pH value, and NH4+-N concentration in surface water. Six levels of reed (Miscanthus lutarioriparius) biochar amount, i.e., 0%, 1%, 2%, 4%, 6% and 8% of soil weight of the 0-20 cm column, were applied in two typical paddy soils in sou-thern China, i.e., reddish clayey soil derived from quaternary red soil and granitic sandy soil derived from granite. Compound fertilizer was applied at a rate of 200 kg N·hm-2. The results showed that biochar application resulted in significant differences in the rate and cumulative amount of ammonia volatilization between the two soils and among different biochar treatments. For the granitic sandy soil, peak ammonia volatilization under different biochar treatments appeared at the second day after fertilization, which was decreased by 23.6%-53.4%. For the reddish clayey soil, peak ammonia volatilization appeared between the 7th to 13th day after fertilization, which increased with biochar addition level. The rate of ammonia volatilization from the granitic sandy soil was generally higher than that from the reddish clayey soil. For the granitic sandy soil, addition of <4% biochar could inhibit the ammonia volatilization and cumulative volatilization amount, with the greatest reduction (46.9%) at the treatment with 2% biochar addition. The addition of biochar did not affect the pH value of surface water at the early stage of rice growth. For the reddish clayey soil, the pH value and NH4+-N concentration in the surface water increased with biochar addition level, resulting in the increases of ammonia volatilization rate and cumulative volatilization amount by 1.3-10.5 times. Biochar addition level was the key factor affecting ammonia volatilization from the two soils. Elovich equation could well fit the variation trend of cumulative ammonia volatilization with time for the two soils, with the correlation reaching extremely significant level for each treatment. Overall, the application of reed biochar could suppress ammonia volatilization from the granitic sandy soil which was nearly neutral in acidity, while would promote ammonia volatilization via increasing pH value and NH4+-N concentration of surface water for the acidic reddish clayey soil. Therefore, appropriate dosages of reed biochar should be selected for different types of soil in order to reduce nitrogen loss.

Vascular inflammation could occur in all organs and tissues in patients with systematic lupus erythematosus (SLE), of which skin is the most frequent one. Our previous research identified anti-galectin-3 (Gal3) antibodies (Abs) as an important mediator of lupus cutaneous vasculopathy. Herein, we showed that anti-Gal3 Abs dysregulated the function of vascular endothelial cells with higher transcript levels of IL-1ß and increased expression of mature IL-1ß. The enhanced production of IL-1ß secreted by endothelial cells was dependent on NLRP3 inflammasome. Intradermal injection of anti-Gal3 Abs in mice induced local inflammation with perivascular infiltration of T cells and neutrophils, which was inhibited by IL-1ß blockade. Induction of anti-Gal3 Abs in circulation by immunization of Gal3 antigen not only led to histopathologic changes in the skin, including focal keratinocytes vacuolization and thickening of blood vessels, but also a systemic autoimmune phenotype that involves autoantibody production and kidney damage. Intriguingly, local overexpression of IL-1ß was primarily associated with skin lesions but not with other internal organs in mice. Finally, we showed that the serum levels of IL-1ß were comparable between SLE patients and healthy donors. Whilst the expression of IL-1ß was enriched in local area with perivascular inflammation in lupus skin lesion compared to healthy normal skin. The results strongly suggest that IL-1ß plays an important role in mediating anti-Gal3 Ab-induced skin vascular inflammation and raised the prospect for using IL-1ß blocking therapies to treat lupus cutaneous damage.

Psoriatic arthritis (PsA) is a unique immune-mediated disease with cutaneous and osteoarticular involvement. However, only a few studies have explored the susceptibility of osteoarticular involvement in psoriasis (Ps) at the genetic level. This study investigated the biomarkers associated with osteoarticular participation and potential shared molecular mechanisms for PsA and ankylosing spondylitis (AS). Methods: The RNA-seq data of Ps, PsA, and AS in the Gene Expression Omnibus (GEO) database were obtained. First, we used the limma package and the weighted gene co-expression network analysis (WGCNA) to identify the potential genes related to PsA and AS. Then, the shared genes in PsA and AS were performed using the GO, KEGG, and GSEA analyses. We also used machine learning to screen hub genes. The results were validated using external datasets and native cohorts. Finally, we used the CIBERSORT algorithm to estimate the correlation between hub genes and the abundance of immune cells in tissues. Results: An overlap was observed between the PsA and AS-related modules as 9 genes. For differentially expressed genes in AS and PsA, only one overlapping gene was found (COX7B). Gene enrichment analysis showed that the above 9 genes might be related to the mRNA surveillance pathway. The GSEA analyses showed that COX7B was involved in adaptive immune response, cell activation, etc. The PUM1 and ZFP91, identified from the support vector machine, had preferable values as diagnostic markers for osteoarticular involvement in Ps and AS (AUC > 0.7). Finally, CIBERSORT results showed PUM1 and ZFP91 involvement in changes of the immune microenvironment. Conclusion: For the first time, this study showed that the osteoarticular involvement in psoriasis and AS could be mediated by the mRNA surveillance pathway-mediated abnormal immunologic process. The biological processes may represent the cross talk between PsA and AS. Therefore, PUM1 and ZFP91 could be used as potential biomarkers or therapeutic targets for AS and Ps patients.

Metal wires have been widely used as substrates for solid-phase microextraction (SPME) fibers instead of commonly fragile silica fibers, but complicated coating modification of their surface is required. Herein, a series of brass wires were soaked in an acidic iron trichloride solution with ultrasonication, which etched the brass surface through a redox reaction. The surface wettability of the pristine brass wire was transformed from hydrophobic to hydrophilic owing to the formation of micro/nanoscale hierarchical structures. After modification with n-octadecanethiol (ODT) and 2-naphthalenethiol (NT), respectively, both wires exhibited superhydrophobicity. Characterization of the resulting wires was conducted using SEM and EDS, and the surface wettability was measured by a contact angle goniometer using identical brass meshes. To build an in-tube SPME-high-performance liquid chromatography (HPLC) online system, the extraction tube was connected with HPLC equipment by replacing the sample loop of a six-port valve. Four types of wires, including the pristine hydrophobic brass wire, the hydrophilic wire after chemical etching, and both superhydrophobic wires, were comparatively applied to the extraction of six estrogens. The optimized extraction conditions were a sample volume of 60 mL, an injection rate of 2 mL/min, and a desorption time of 2 min at a flow rate of 1 mL/min. The results showed that the highest estrogen extraction efficiency was obtained using the superhydrophobic wire modified by NT, with the enrichment factors in the range of 36-350. Furthermore, the superhydrophobic NT wire exhibited a higher extraction efficiency than the ODT wire with identical superhydrophobicity. This demonstrated that the higher extraction efficiency was mainly dependent on  π-π interactions between the sorbent containing naphthalene rings and the target compounds containing benzene rings, rather than surface wettability.
